A phenotypic hallmark of cancer is aberrant transcriptional regulation. Transcriptional regulation controlled by a complicated array molecular factors, including the presence transcription deposition histone post-translational modifications, and long-range DNA interactions. Determining identity function these various factors necessary to understand specific aspects biology reveal potential therapeutic targets. Regulation genome typically studied using chromatin immunoprecipitation followed sequencing (ChIP-Seq) that identifies genome-wide binding interactions through use factor-specific antibodies. long-standing goal in many laboratories has been development ‘reverse-ChIP’ approach identify unknown partners at loci interest. variety strategies have employed enable selective biochemical purification sequence-defined regions, single-copy loci, subsequent analytical detection associated proteins. This review covers mass spectrometry techniques quantitative proteomics before providing survey approaches toward for sequence-specific as technique. fully realized reverse-ChIP technique holds great identifying cancer-specific targets personalized regimens.
